Mutant Allele Imbalance in Cancer

چکیده
The search for somatic mutations that drive the initiation and progression of human tumors has dominated recent cancer research. While much emphasis been placed on characterizing prevalence function driver mutations, comparatively less is known about their serial genetic evolution. Indeed, study this phenomenon largely focused tumor-suppressor genes recessive at cellular level or mechanisms resistance in with mutant oncogenes targeted by therapy. There is, however, a growing appreciation despite decades-old presumption heterozygosity, changes oncogene zygosity are common dosage stoichiometry lead to selective growth advantages. Here, we review progress understanding allele imbalance its implications tumor biology, evolution, response anticancer

KRAS mutant allele-specific imbalance (MASI) assessment in routine samples of patients with metastatic colorectal cancer.
AIMS Patients with colorectal cancer harbouring KRAS mutations do not respond to antiepidermal growth factor receptor (anti-EGFR) therapy. Community screening for KRAS mutation selects patients for treatment. When a KRAS mutation is identified by direct sequencing, mutant and wild type alleles are seen on the sequencing electropherograms. KRAS mutant allele-specific imbalance (MASI) occurs when...
متن کاملAllele-specific p53 mutant reactivation.
Rescuing the function of mutant p53 protein is an attractive cancer therapeutic strategy. Using the National Cancer Institute's anticancer drug screen data, we identified two compounds from the thiosemicarbazone family that manifest increased growth inhibitory activity in mutant p53 cells, particularly for the p53(R175) mutant. Mechanistic studies reveal that NSC319726 restores WT structure and...
متن کاملOncogene Mutations, Copy Number Gains and Mutant Allele Specific Imbalance (MASI) Frequently Occur Together in Tumor Cells
BACKGROUND Activating mutations in one allele of an oncogene (heterozygous mutations) are widely believed to be sufficient for tumorigenesis. However, mutant allele specific imbalance (MASI) has been observed in tumors and cell lines harboring mutations of oncogenes. METHODOLOGY/PRINCIPAL FINDINGS We determined 1) mutational status, 2) copy number gains (CNGs) and 3) relative ratio between mu...
متن کاملCDKN1C expression in Beckwith-Wiedemann syndrome patients with allele imbalance.
In this study, we have examined CDKN1C expression in BWS patients with allele imbalance (AI) affecting the 11p15 region. Two of two informative patients with AI, attributable to mosaic paternal isodisomy, exhibited reduced levels of CDKN1C expression in the liver and kidney, respectively, relative to expression levels in the equivalent tissues in normal controls. Although overall expression was...
متن کاملDelimiting Allelic Imbalance of TYMS by Allele-Specific Analysis
Allelic imbalance of thymidylate synthase (TYMS) is attributed to polymorphisms in the 5'- and 3'-untranslated region (UTR). These polymorphisms have been related to the risk of suffering different cancers, for example leukemia, breast or gastric cancer, and response to different drugs, among which are methotrexate glutamates, stavudine, and specifically 5-fluorouracil (5-FU), as TYMS is its di...
